PROJECT TITLE :

Electrospun scaffold containing TGF-β1 promotes human mesenchymal stem cell differentiation towards a nucleus pulposus-like phenotype under hypoxia

ABSTRACT:

The study was geared toward evaluating the result of electrospun scaffold containing TGF-βone on promoting human mesenchymal stem cells (MSCs) differentiation towards a nucleus pulposus-like phenotype underneath hypoxia. 2 sorts of nanofibrous scaffolds containing TGF-βone were fabricated using uniaxial electrospinning (Group I) and coaxial electrospinning (Cluster II). Human MSCs were seeded on both sorts of scaffolds and cultured during a hypoxia chamber (a pair ofp.c O2), and then the scaffolds were characterised. Cell proliferation and differentiation were also evaluated after three weeks of cell culture. Results showed that each kinds of scaffolds shared similar diameter distributions and protein release. But, Group I scaffolds were a lot of hydrophilic than that of Cluster II. Both types of scaffolds induced the MSCs to differentiate towards the nucleus pulposus-type phenotype in vitro. Additionally, the expression of nucleus pulposus-associated genes (aggrecan, kind II collagen, HIF-oneα and Sox-nine) in Group I increased additional than that of Cluster II. These results indicate that electrospinning nanofibrous scaffolds containing TGF-β1 supports the differentiation of MSCs towards the pulposus-like phenotype in an exceedingly hypoxia chamber, that would be a more appropriate selection for nucleus pulposus regeneration.
